Dr. Li is widely recognized for his pioneering contributions at the interface of immune regulation, proteostasis, and translational immuno‑oncology. Over the past two decades, his laboratory has established key principles linking endoplasmic reticulum (ER) proteostasis to immune function. His team was among the first to apply mammalian genetics to elucidate the in vivo roles of GRP
94/gp96
, an ER‑resident HSP
90 paralog, demonstrating its function as a client‑restricted, ATP‑dependent immune chaperone essential for the organization of Toll‑like receptors, integrins, Wnt co‑receptors, the GARP–TGF‑β axis, and platelet biology. These discoveries helped launch the field of immune chaperone biology and have produced multiple translational innovations, including gp96‑based cancer vaccines and first‑in‑human anti‑GARP CAR‑T therapies
.
