Scientist, Computational Protein Engineering & Design About Tectonic Therapeutic:
Tectonic is a clinical stage biote chnology company focused on the discovery and development of therapeutic proteins and antibodies that modulate the activity of G-protein-coupled receptors (GPCR). Tectonic’s founders are at the forefront of scientific research in biochemistry and molecular pharmacology. Their extensive track record of achievements includes major contributions to the development of multiple approved therapies for the treatment of cancer, autoimmune diseases and beyond.
Job SummaryTectonic Therapeutic is seeking a PhD-level Scientist with deep expertise in protein biochemistry, structural biology and computational biology. This role will focus on the computational design and optimization of therapeutic biologics, with a particular emphasis on leveraging state-of-the-art algorithms to advance GPCR-targeted antibody discovery. The candidate should have strong hands‑on experience applying machine learning, AI, and deep learning methods to protein and antibody engineering.
You will operate as a "power user" of modern computational tools, balancing the speed of generative AI with the rigor of physics‑based modeling to deliver high‑quality biologics leads.
- Lead Generation via Generative AI: Deploy, adapt, and critically evaluate state‑of‑the‑art machine learning and deep learning models (e.g., protein language models, structure‑based diffusion models, and related generative frameworks) for the design of novel antibody candidates against GPCR targets.
- Computational Strategy & Model Integration: Partner with the Director of Computational Biology to select, implement, and evolve computational and AI/ML approaches that enhance GPCR antibody discovery workflows, including model selection, benchmarking, and integration with experimental data.
- Structural Interface Analysis: Perform detailed structural characterization of antibody‑antigen interfaces. You will identify key residue interactions, map epitopes, and ensure that designed paratopes are sterically and chemically compatible with the target.
- Physics‑Based Refinement: Apply physics‑based principles to validate and refine AI‑generated designs. This includes conducting energetic scoring and performing atomistic refinement to ensure biophysical stability.
- Multi‑Objective Optimization: Optimize antibody leads for both affinity and "develop ability." You will balance binding strength with crucial biophysical traits like solubility, thermal stability, and low aggregation propensity.
- Iterative Design‑Test Cycles: Work closely with experimental teams to integrate assay data back into your computational workflows. Use wet‑lab feedback to troubleshoot, retrain, and refine computational and machine learning models, iteratively improving predictive performance and design quality.
- Education: PhD in Computational Biology, Bioinformatics, Biophysics, Biochemistry, Structural Biology, or a related quantitative field.
- Biochemical Foundation: Strong understanding of the physical chemistry of proteins, amino acid interactions, and the thermodynamics of protein folding/binding.
- Computational Biology Expertise: Demonstrated ability to design, manipulate, and analyze large‑scale biological and structural datasets, and to translate algorithmic outputs into actionable biological insights.
- AI/ML Application: Strong hands‑on experience applying, adapting, and interpreting machine learning and deep learning models for protein or antibody design (e.g., structure prediction, sequence design, or representation learning).
- Physics‑Based Toolkit: Proficiency in molecular mechanics, force‑field‑based scoring, and structural refinement protocols.
- Programming: Proficiency in Python for data analysis, automation of structural pipelines, and handling PDB/sequence data.
